Huanglongbing (HLB), associated with a non-culturable bacterium ‘Candidatus Liberibacter asiaticus’ (CLas), is a highly destructive citrus disease with a long but poorly documented history in China. No effective treatment for HLB is available. The identification of new prophages in abundant CLas genomic sequence data provides new insights into both the diversity of CLas strains and HLB management. In this study, CLas populations from nine provinces were surveyed for the presence of prophage. Two major prophage typing groups (PTGs) were discovered to be associated with two different altitude regions: strains of CLas in PTG1 from high altitude regions (HAR) mainly contained prophage Type 1 only or Types 1 and 3, whereas strains of CLas in PTG2 from low altitude regions (LAR) mainly contained prophage Type 2. The discovery of these CLas population patterns provides evidence for independent origins of HLB in HAR and LAR. Guangdong province is the generally recognized domestic region of origin for HLB and is primarily responsible for the dissemination of HLB in LAR through transport of seedlings. Both Yunnan and Sichuan provinces are the probable regions of origin for HLB in HAR. PTG2 was further divided into two subgroups: PTG2-1, found in Guangdong, Fujian and Guangxi and PTG2-2, found in Jiangxi, Zhejiang and Hunan. These regions and prophage types are correlated with early and late introductions of HLB in LAR. Eight experiments were carried out in Denmark to determine the yield loss of spring barley due to Cirsium arvense in farmers' fields and to suggest and evaluate a novel approach for quantifying C. arvense infestation in large plots. Literature about the competitive ability of C. arvense is old, scattered and inconclusive, and existing models for estimating crop yield loss are based on data from North America. This study showed that C. arvense coverage could be quantified from unmanned aerial vehicle imagery using a manual image analysis procedure. This gave similar results as scoring the coverage. Yield loss of spring barley due to C. arvense infestation assessed at harvest was given by Y = 100·(1−exp(−0.00170·X)) where Y is the percentage of crop yield loss and X is the percentage of C. arvense coverage. The yield loss was much lower than estimates from models that have been developed in North America. It is speculated that the main reason for this is the later emergence of C. arvense than the crop due to lower soil temperatures in spring. Grain moisture increased linearly with C. arvense coverage: M = 0.0310·X where M is the proportional (%) increase in grain moisture and X is the proportion (%) of C. arvense coverage. Automated image analysis procedures are needed to estimate C. arvense coverage on field scales, and further experiments are needed to reveal whether the low competitive ability of C. arvense found in this study is representative for Northern Europe. 相似文献

The application of disinfectants through drip irrigation could be a feasible practice against verticillium wilt (Verticillium dahliae) of olive. OX-VIRIN (activated peroxide) and OX-AGUA AL25 (quaternary ammonium compounds) are two disinfectants that have shown efficacy against V. dahliae in irrigation water and potential for reducing the disease in young olive plants. In this work, various post-planting application strategies incorporating OX-VIRIN (once a month, or twice a month on alternate or successive weeks) or OX-AGUA AL25 (once a month, or twice a month on alternate weeks) were assessed for their effect on V. dahliae in soil, disease in olive trees, and olive yield, in a 2-year pot-experiment under natural environmental conditions. The disinfectants were injected via metering pumps into a drip irrigation system that irrigated olive trees planted in V. dahliae-inoculated soil. All the application strategies significantly reduced the total inoculum density in soil compared to controls with no disinfectants and noninoculated soil. The microsclerotia density was also significantly reduced in disinfested soils by 73.6–86.8%, depending on the strategy. The symptoms and infection incidence were always lower in treatments subjected to disinfestation. The treatment with OX-AGUA AL25 applied twice a month on alternate weeks most reduced the symptoms (by 53.0%) and colonization index (by 70.8%) with respect to untreated water control. This soil disinfestation also significantly strengthened the symptom remission. Tree growth and production were negatively affected by soil inoculation (reduced by 45.6% and 88.7%, respectively), but not so by disinfectants, which even relieved the reduction in inoculated soils, especially when OX-AGUA AL25 was applied. 相似文献

Development of common bean cultivars with partial white mould resistance through breeding techniques has been a challenge in Brazil. As yet, lines/cultivars from breeding programmes have not been investigated for resistance; therefore, this study screened 107 lines/cultivars for their reactions to white mould in 14 preliminary trials conducted under irrigation. Thirteen resistant lines/cultivars (three of Andean origin) and six Mesoamerican cultivars (three intermediately resistant and three susceptible) were selected for further investigation. These lines/cultivars and the resistant control A195 were evaluated in six advanced trials and two straw tests to assess the effectiveness of the screening procedure. In 11 preliminary trials, screenings were performed under moderate/high or higher disease pressure. These pressures occurred in two advanced trials in which, when yields were averaged across moderate/high and high pressures, 10 Mesoamerican lines/cultivars selected for resistance yielded 14%, 23%, and 38% more than intermediately resistant cultivars, A195, and susceptible cultivars, with median disease ratings (1–9 scale) of 4.5, 5.7, 5.7, and 6.7, respectively. In the straw test, three Andean lines/cultivars (A195 included) and two susceptible cultivars in the field were among those with the highest levels of physiological resistance. Thus, field rating under high disease pressure and greenhouse rating did not correlate significantly, suggesting that field trials are critical to evaluating resistance and to identifying high-yielding beans. Therefore, lines/cultivars from breeding programmes assessed in field trials may provide a low cost and fast way to identify high-yielding bean cultivars with partial resistance to white mould in the subtropical southern hemisphere. 相似文献

The cotton mealybug, Phenacoccus solenopsis Tinsley 1898 (Hemiptera: Pseudococcidae), is recorded from Saudi Arabia for the first time. This species was collected from heavily infested Hibiscus rosa-sinensis L. (Malvaceae) from the city of At-Taif in the south-western part of the Arabian Peninsula. In addition, the mealybug Ferrisia virgata (Cockerell, 1893), previously recorded in Saudi Arabia, was found on Plumeria rubra L. (Apocynaceae) and on Dodonaea viscosa Jacq. (Sapindaceae). Slide mounts were prepared and voucher specimens were preserved at the University of Jordan Insects Museum. Further surveys should be initiated to investigate the distribution and hosts of P. solenopsis in Saudi Arabia. 相似文献

Photosynthesis is one of the most important biological reactions and forms the basis of crop productivity and yield on which a growing global population relies. However, to develop improved plant cultivars that are capable of increased productivity, methods that can accurately and quickly quantify photosynthetic efficiency in large numbers of genotypes under field conditions are needed. Chlorophyll fluorescence imaging is a rapid, non-destructive measurement that can provide insight into the efficiency of the light-dependent reactions of photosynthesis. To test and validate a field-deployed fluorescence imaging system on the TERRA-REF field scanalyzer, leaves of potted sorghum plants were treated with a photosystem II inhibitor, DCMU, to reduce photochemical efficiency (FV/FM). The ability of the fluorescence imaging system to detect changes in fluorescence was determined by comparing the image-derived values with a handheld fluorometer. This study demonstrated that the imaging system was able to accurately measure photochemical efficiency (FV/FM) and was highly correlated (r = 0.92) with the handheld fluorometer values. Additionally, the fluorescence imaging system was able to track the decrease in photochemical efficiency due to treatment of DCMU over a 7 day period. The system’s ability to capture the temporal dynamics of the plants’ response to this induced stress, which has comparable dynamics to abiotic and biotic stressors found in field environments, indicates the system is operating correctly. With the validation of the fluorescence imaging system, physiological and genetic studies can be undertaken that leverage the fluorescence imaging capabilities and throughput of the field scanalyzer. 相似文献

Antimicrobial stewardship (AMS) is the term increasingly used to describe the multiple approaches needed to sustain the efficacy of antimicrobial drugs in the face of the increasing development and spread of antimicrobial resistance in bacterial pathogens, and the global crisis in medicine that it is engendering. The concept and the practices associated with AMS continue to evolve but the general approach is a dynamic and multifaceted one of continuous improvement based on reducing, improving, monitoring and evaluating the use of antimicrobials so as to preserve their future efficacy and to protect human and animal health. Using many equine examples, this basic overview discusses the multiple and interacting elements of AMS: Practice guidelines, infection control and prevention, clinical microbiology, resistance and use surveillance, dosage, pharmacokinetics and pharmacodynamics, regulation, education and owner compliance, leadership, coordination and measurement. There have been impressive advances in recent years in reporting and analysis of AMR in horses, in the scrutiny and assessment of how antimicrobial drugs are used in horses and in identification of areas for improvement including dosing, surgical prophylaxis, infection control, development of practice standards and the use of clinical microbiology. Antimicrobial stewardship is taking shape as we start to see the emergence of evidence-based recommendations but far more is required. Containing and even rolling back AMR will need the continued engagement of practitioners, equine national and international practitioner organisations, researchers and educators in the academic community, horse owners, regulators and others. 相似文献

The change in soil carbon (C) concentration, soil pH and major nutrients for approximately 1,000 topsoil sampled from on-farm experimental sites over a thirty-year period from 1950 to 1980 in north-east Scotland are summarized. This period coincided with increased agricultural intensification, which included regular liming and fertilizer additions. During 2017, 37 of these sites were resampled and reanlaysed. While pH and extractable phosphorus (P) and potassium (K) increased over this period, there was no detectable change in the percentage loss on ignition. Composite soil samples were taken by auger from a depth of 0–15 cm and compared with the corresponding archived samples collected at the initiation of each experiment. Analysis of these resampled soils indicated no significant change in soil carbon (C), although soil pH, extractable magnesium (Mg) and K and Nitrogen (N) concentrations were significantly greater (p < .001) but extractable soil P concentration was significantly less (p = .015) compared with the original samples. Even though measuring C concentration alone is a poor indicator of overall changes in soil C stocks, it does provide a relative quick “early warning” of C losses that would justify a more comprehensive measure of stocks. 相似文献

Calonectria leaf blight, caused by Calonectria pteridis, is currently one of the main foliar diseases in eucalypt plantations in Brazil. In warm and high rainfall regions, the disease can be a limiting factor for eucalypt production when planting susceptible genotypes. The most effective method for controlling this disease in the field is the use of resistant genotypes, which requires knowledge of the genetic variability and aggressiveness of the pathogen population for effective deployment of plant resistance. This work evaluated the genetic diversity and aggressiveness of C. pteridis populations obtained from infected eucalypt plants in Monte Dourado (Pará state) and Imperatriz (Maranhão state), Brazil. To study the genetic diversity, 16 ISSR primers were tested, five of which amplified polymorphic, reproducible and informative bands. Thirty-one closely related genotypes were identified from 84 isolates studied, indicating that the population has a low genetic diversity. The aggressiveness of seven isolates, selected according to geographic origin and their clustering in the ISSR-based dendogram, was determined by inoculation of a hybrid Eucalyptus grandis × E. urophylla clone under controlled conditions. Disease severity was assessed by both measuring the percentage of plant defoliation and assigning a score according to a diagrammatic scale of symptoms. A high correlation between the two evaluation methods was observed, which revealed significant differences in aggressiveness among the isolates. The diagrammatic scale is recommended for disease evaluation because results are obtained much faster, before the occurrence of severe defoliation. No correlation between clustering in the ISSR-based phylogenetic analysis and aggressiveness was observed. 相似文献

The effect of three mono‐specific and four combinations of the diatoms Chaetoceros muelleri, Chaetoceros calcitrans, and the chlorophyte Tetraselmis suecica on survival, development, and dry weight of the western school prawn, Metapenaeus dalli, was assessed from protozoea I until mysis I. The development and dry weight of larvae were significantly greater when fed diets comprising C. muelleri and/or T. suecica. A fourth diet, consisting of all three microalgal species also performed just as well. Survival alone was a poor measure of the performance of the various diets. Larvae fed with C. calcitrans, either alone or in a mixed diet with either C. muelleri or T. suecica, had significantly slower development and lower dry weight. Overall performance, assessed using the normalized biomass development index, determined that both mono‐specific and mixed diets containing C. muelleri and T. suecica were among the best for M. dalli larvae. These results for M. dalli are consistent with those found for commercially grown penaeid prawns. This study enhances the limited knowledge on the feeding requirements of metapenaeid prawn larvae. Moreover, the results will help improve hatchery methods for the aquaculture‐based enhancement of M. dalli in the Swan‐Canning Estuary and potentially increase the abundance of this iconic recreational species. 相似文献
